When embarking on a long-distance relationship LDR and each party has gone past the stage of being crazy for one another but have yet to meet in person. Normally this would be called 'fucked' but because you've not had the chance to fuck yet, you're 'pre-fucked' or 'prefucked'. It is a portmanteau, or blend, of the words "perfect" and "fucked". The meaning depends on the context it's used in:

Completely ruined or messed up: This is the most common meaning. Combining "perfect" (as in, complete or total) with "fucked" results in a phrase that conveys a state of being utterly and totally screwed or destroyed.
Ironic or sarcastic usage: The word could be used with heavy irony to describe a situation that is the exact opposite of perfect.

Emphasis on the first syllable which is drawn out hence the second letter being a "u".
