Driving a vehicle (particularly the engine) hard for a few minutes, getting everything fully warmed up and loaded, for the purpose of improving its state of running. Has some theoretical and empirical validity to it (in certain cases, it does actually work), but it can also be an incompetent hackjob substitute for actual mechanical skills and effort (in these cases, it usually makes things worse).
An Italian tuneup is the machinery getting exercise.

A minor (or major) beat down of a guy deficient in payment of loansharking or protection payments to the 'mob' or who displays a lack of respect to same.
"Joey the deli guy is a little slow on the payback and he gave Vinnie some backtalk the last time he collected. I think it's time Joey got a little tuneup to remind him who's boss in this town."
