University of Tears comes from University of Toronto in Canada. University of Toronto is highly competitive and workload are heavy. Everyone has tears every day. Some courses are too heavy that could compare to Harvard or Yale.

It is a show that a beast, no matter how savage, has a heart that is capable of feeling. A Manly Tear (so awesome it needs to be capitalized) demonstrate the control, and the precision of a man's character, as he is willing to skirt the lines of tasteful shows of emotion and uncontrollable babbling. Lesser men are incapable of such restraint, and therefore cannot shed a Manly Tear.

In episode 26, the main bad guy, Vicious (after his failed coup against his employer) confronts his final boss and tells him, "Now you will cry Scarlet Tears." He then places his sword on the boss's nose at eye level, and cuts his boss's eyes, simultainously killing him. Vicious then takes control of their criminal empire.
A very memorable phrase in anime.
