This colloquial phrase is derived from the root of eating - where one must have "their napkins ready" as if they are having a meal. Therefore, having one's napkin ready means they will be adequately prepared for the event and/or activity (e.g. having a round of drinks, entering a club or bar, playing a sport, engaging in sexual activity, etc)
