This is a catch phrase that originated in a TV ad in the late Eighties that featured super athlete extraordinaire Bo Jackson and rock'n'roll pioneer Bo Diddley (he's notorious for his famous box-shaped electric guitar). Basically, the term means that somebody doesn't know doodly squat.
The narrator says:
Bo knows football.
Bo knows baseball.
Bo knows weightlifting.

Bo, you don't know diddley!

Get the Don't quit your day jobmug. This is term most commonly used as a statement urging someone to get revenge. It is often used when one is feeling especially vengeful for this certain wrong.
Gandalf: Gimli just broke into my house and ate all of my cupcakes, you know, the ones with the little hearts.
Bilbo: Oh?
Gandalf: And he shaved my beard, i'm really mad.

Get the Don't get mad, get even.mug. A phrase used to ridicule someone's stupidity, "big white mint" refers to a urinal cake. Thus "don't eat the big white mint" implies that someone is stupid enough to mistake the urinal cake for a mint and eat it.
I was on a date with this really dumbass guy, and he said he had to go to the bathroom. I said, OK, don't eat the big white mint.
