A person who is "marked" has enemies after them. In older English, we would say they are a "wanted man" or "being hunted", meaning that there is a person or group of people actively seeking out that person, with a desire to ruin their life or injure them. More commonly however, it is used ironically to describe a situation when something bad happens to someone, such as losing something or failing at a simple task.
